Automatic air vent screw

Just a quick question the black screw on the vent is it suppose to be screwed tight or undone under normal operation

Forgot to say a main combi boiler

Its meant to be slightly loose, to allow air to escape. However, as they nearly always leak after a while you often find them closed.
